Pooled Fish Hepatocytes – a new in-vitro model System.
Animals like Atlantic salmon (Salmo salar) have been used in vivo by researchers for decades to assess bioaccumulation, metabolism, and environmental risk of chemicals, including drugs, xenobiotics, cosmetics, fragrances, flavouring substances, herbicides and crop protection products. More recently, in vitro studies have demonstrated that hepatocytes, microsomes, and hepatic S9 fractions are effective and efficient tools to predict the in vivo endpoint for biotransformation and accumulation testing.
PRIMACYT is one of the pioneers in the development of advanced in vitro model systems. We are a leading company in the field of research of human and animal hepatocytes. We offer fresh and cryopreserved animal hepatocytes in suspension and plated.
PRIMACYT is approved by the European Commission as an EU Reference Laboratory for Alternatives to Animal Testing (EURL ECVAM) since 2013 and serves as member of the laboratory network responsible for the method validation.
Our colleagues are professionals and undergo continuous training in the preparation, isolation and cryopreservation of hepatocytes in general, including related co-products, methods and tools. We are a GLP certified laboratory since 2006.
Animal Tissue Performance and Regulations. The acquisition of animal tissue is performed under permits 3902-046-2017 and DE 13 076 0001 21 from the Landkreis Ludwigslust-Parchim, Department of Veterinary- and Foodstuff Control, following EU Regulation No. 1029/2009, Art. 17 and 23 and permit 7221.3-2-004/17 from the Ministry for Agriculture, Food Safety and Fishery of the State Mecklenburg-Pomerania in accordance with EU Regulation 2010/63/EU, and with several additional regulations for the protection of animals (e.g., TierSchG dated July 18, 2016, and TierSchVerV dated August 31, 2015) used for experimentation and other scientific purposes.

The animals were kept under controlled housing conditions in compliance with the OECD guideline 319A established for rainbow trout.
PRIMACYT offers certified pools of cryopreserved salmon hepatocytes. Pooled hepatocytes were obtained from 3 to 8 individual animal liver perfusions performed in parallel.
Each pool was comprehensively tested for viability, plateability and use in suspension. Cytochrome P450/Phase I and Phase II activities were determined to characterise the products.
