PRIMACYT can provide researchers with Beagle tissue panels, FDA (Federal Drug Administration) conform. Tissue panels form dogs can be provided either as frozen tissue blocks or formalin fixed and paraffin embedded (FFPE).
There are two major tissue preparation methods used today. Tissues can be saturated with formalin and then embedded in a block of paraffin wax (referred to as formalin-fixed paraffin-embedded or FFPE tissue). Alternatively, it has become feasible to “flash-freeze” tissue and store the frozen tissues at below -80°C in ultra-low freezers. Each method comes with advantages and disadvantages.
Formalin-fixed paraffin-embedded (FFPE) tissues are the most common source of archived material. FFPE tissues are used for immunohistochemical analysis mainly in oncology, the study cancer of cancer, hematology to study blood disorders, bone marrow disorders, immunology and for comparative analysis of healthy vs. diseased tissues.
Compared to frozen tissues, FFPE tissues has several advantages and disadvantages:
Advantages:
- Cost effective and simple storage at room temperature
- Easy biobanking
- Stable for many years
- Formalin and wax preserve fragile structures inside and between the cells
- Preservation of proteins allows for immunohistochemical analysis
Disadvantages:
- Formalin is volatile and toxic
- Paraffin embedding and fixation procedures may be time consuming
- proteins may be denatured and therefore may not bind to the same antibodies as the native (not-denatured) proteins
- DNA and RNA are not very well preserved: may not be suitable for molecular genetic analysis such as PCR, qPCR or next generation DNA sequencing
Major applications for frozen tissues are molecular genetics analysis and native morphology studies in oncology, hematology and immunology. Compared to FFPE tissues, frozen tissues has several advantages and disadvantages:
Advantages:
- Easy to prepare
- Useful for immunohistochemical analysis as proteins are preserved in native state
- Suitable for molecular genetic analysis, e. g. PCR, qPCR and next generation DNA sequencing
Disadvantages:
- Ultra-low temperature freezers are required as tissues do not store well in -20°C freezer and degrade quickly at room temperature
- Biobanking is energy consuming and vulnerable to power or mechanical failure or carelessness in the lab
